Few cells conform to the stereotype of the spherical blob hastily scribbled on chalkboards and, regrettably, sometimes even displayed prominently in textbooks. Instead, real cells display a remarkable degree of structural and functional asymmetry. In modern cell biological parlance, this asymmetry has come to be lumped under the general heading of "cell polarity". Cell polarity is by no means restricted to the cells of tissues and organs, but can also be displayed by cells that lead a more solitary existence. The amazing extent to which cell morphology is correlated with function has long been a source of inspiration for biologists. Today the fascination continues unabated in the field of cell polarity, where it is fueled by an ever-deepening appreciation for the ways that fundamental cellular processes, such as membrane trafficking and cytoskeleton assembly, contribute to the establishment and maintenance of cell polarity. Selective cytoplasmic organelle and protein targeting has long been thought to constitute the sole determinant of cell polarity and complexity. This view has been changed, however, by the discovery of differential subcellular RNA distribution patterns. Over the past decade it has become clear that selective mRNA sorting and translation contributes to an accumulation of cognate proteins at discrete cellular sites. In this book, various systems are discussed (e.g. Xenopus oocytes, mammalian brain, invertebrate nervous system) in terms of RNA trafficking, RNA-targeting sequences (cis-elements) and RNA-transporting proteins (trans-factors), activity-dependent translational regulation and the significance of the cytoskeleton for neuronal function and plasticity. It also discusses nucleocytoplasmic export of mRNA and viral RNA as another example of subcellular RNA kinesis.

​​​​​​​Prominin-1 or otherwise known as CD133 is a glycoprotein that is present in humans and mice. Since the first description of prominin in 1997, in mouse neuroepithelial cells and in human hematopoietic stem cells as AC133 antigen, this molecule has aroused a large interest especially, as a stem cell marker, that gave rise to an ever growing body of publications and more recently its expression in cancer stem cells. Controversies as to its role as a cancer stem and its detection in different models, as well as its use as a prognostic marker have emerged. Yet, beyond its use as a stem cell and cancer stem cell marker, prominin-1/CD133 displays unique biological features and appears of importance in other processes like for example in retinal biogenesis. Indeed, this five-transmembrane plasma membrane glycoprotein, which marks membrane protrusions is associated with several essential processes like cell polarity, asymmetric cell division and membrane remodeling. We propose to review current knowledge about this intriguing molecule and present pertinent information to determine the biological role of prominins and assess their importance in medicine and cancer research. The primary audience for this book is geared towards scientists and researchers with interest in cancer stem cells, stem cells, cell biology, neurobiology, and regenerative medicine.

A fundamental characteristic of metazoans is the formation of a simple, polarized epithelium. In higher animals, the structural integrity and functional polarization of simple epithelia require a cell-cell adhesion complex containing a classical cadherin, the Wnt-signaling protein β-catenin and the actin-binding protein α-catenin. I have investigated the evolutionary origins of epithelial cell polarity and of the cadherin-catenin complex. I show that the non-metazoan Dictyostelium discoideum forms a polarized epithelium that is essential for multicellular development. Although D. discoideum lacks a cadherin homolog, I have identified and characterized an α-catenin ortholog that binds a β-catenin-related protein. Both proteins are essential for formation of the epithelium, polarized protein secretion and proper multicellular morphogenesis. Thus the organizational principles of metazoan multicellularity may be more ancient than previously recognized, and the role of the catenins in cell polarity predates the evolution of Wnt signaling and classical cadherins.

Epithelial cells exhibit an apical–basolateral axis of polarity that is generated during embryogenesis, is maintained throughout adult life in the face of constant cell regeneration, and is perturbed in several epithelial-associated diseases. We examine the structural and functional organization of epithelial tissues, as well as the events critical for generating epithelial asymmetry including vectorial trafficking of proteins and lipids, association of signaling and polarity proteins with subdomains of the plasma membrane, and 3D orientation of epithelial cells in response to cell–cell and cell–matrix interactions. As a paradigm to understand how these three processes are coordinated in time and space, we explore apical lumen formation. We also examine the final steps in epithelial morphogenesis, including brush border morphogenesis and ciliogenesis. Finally, we provide examples of disease processes that result from defects in epithelial polarity including diabetes insipidus, microvillar inclusion disease, hereditary deafness, ciliopathies, and cancer.

Protein kinase C (PKC), a family of serine-threonine kinases, rocketed to the forefront of the cancer research field in the early 1980’s with its identification as an effector of phorbol esters, natural products with tumor promoting activity.
